Hello James,
There are two obvious lineages in early Knox County, TN. Both are
identifiable by their Revolutionary War pensions. One lineage has John
Childress born 1759 and the other lineage has a Mitchel Childress born
1750. I have seen a reference to John the third relating to the
descendants of Mitchel Childress. But both lineages have descendants
using the John name.
The John Childress born 1759 has descendants who have taken the DNA test
already. Mitchel Childress born 1750 has one purported descendant in
the process of taking the DNA test. My expectation is that both Mitchel
Childress (b. 1750) and John Childress (b. 1759) are related. The 12
marker DNA test would definitely confirm or disaffirm if you are related
to those who have taken the test or are taking the test. But if both
John (b. 1759) and Mitchel (b. 1750) are related, the DNA test is not
likely to tell you which lineage you are more closely related to,
although it is possible the Mitchel lineage will have a distinctive
mutation when all the pending results, or future test results, are
finally known.
The 12 marker DNA test would show if you shared a common ancestor with
either lineage. The DNA test won't tell you HOW you are related to a
person, only that you are biologically related. Its up to the paper
courthouse records (wills, deeds, census records, etc) to fill in the
details.
I would indeed recommend you take the 12 marker test. It will shed some
more light on your research. Even if your test results says you are in
another group it would be useful even to know that and give direction on
how to interpret records.
Regarding your Jackson County, Alabama question, I have no insights that
come to mind.

You ask: My ancestors came from Trimble Cnty., Ky. Is that near Barren
Cnty.?
Answer: I don't know.... you'd have to have, however, a paper trail
showing your relatives moved from Barren County to Trimble County.
We are trying to find descendants of a Goochland County family...some of
whom went to Barrens County, KY.
I appreciate your enthusiasm. I wish more people were
as eager to find DNA participants as you. I find the lineages I need,
but then the modern descendants don't want to participate.
We are trying to find evidence to test the following hypothesis:
1) Group One DNA results are descendants of the "Childers" families who
immigrated after 1640 including Abraham, Philomen, and Thomas
2) Group Two DNA results are descendants of the "Childress" families who
immigrated in 1745 and are related to the Sarah Childress Polk lineage.
The Sarah Childress Polk lineage left a gravestone that spoke of their
1745 immigration to Virginia. There also appears to be a Goochland
County, VA family that immigrated circa 1755...ten years later...that
may be related to those of the 1745 immigration.
As always, any DNA must come from the Y-chromosome...and only males have
that chromosome. And the male must carry the Childress or Childers
surname or similar spelling to qualify his DNA as descending without
interruption from those first immigrants.
We have over 24 test results completed, 6 more in process, and 4 on the
fence.
Next month I hope we will get some answers about Mitchel Childress, born
1750 in Henrico County, VA and died in Knox County TN. Also on the
fence are another DNA test or two from the Goochland County, VA area.
I am trying to get a descendant of Benjamin Childress of Colonial
Albemarle County, VA to take a DNA test, and also a descendant of Surrey
County, N.C. inhabitants.
