It is very possible for brother's to differ as a mutation can occur at
anytime. My grandson has retinoblastoma which is caused by a mutation and
then passed on to descendents. Neither his older brother, younger sister,
parents, aunts, uncles, grandparents, etc has ever been diagnosed with this.
At the same time, if he and his brother were to have their Y-DNA tested for
genealogy purposes it is just as possible that they would match 37/37 as
not.
We can't get too hung up with using just DNA to determine if someone is a
brother, cousin, 7th great-grandson, etc. If you are a close match and your
genealogy research shows a relationship then you are related and your paper
trail determines what the relationship is, not the DNA numbers. If you are
a close match with someone with a different surname but cannot connect
through genealogy you are still related - it's just that the relationship
probably occurred before the use of surnames and is of no genealogical
value. My dad has so many matches with other surnames it would be
impossible to determine how they are related - it was probably in the UK
prior to 1300 AD.
By the way Ken, are you related to Robert "King" Carter from your male or
female Carter ancestor? If it is the female,Y-DNA testing will not be able
to prove it.

Carters. We know the name of my 3rd great grandfather, Charles Edward
Carter, but the paper trail is elusive.>
The genealogy I have posted goes back to me 2nd grandfather, and from
there
goes back on the maternal side (my 3rd great grandmother) to William
Carter
of Middlesex County (1641).
Our DNA is 2 markers off from Group 2 in the 24 marker test, and 3 markers
distant in the 25 marker test. The 3rd marker difference is the same
marker
that my brother and I differ on.
